PASS does most commonly used power analysis. However, if you are doing a complicated analysis, it's better to have someone trained in statistics to help you, especially for a grant application. Do it by hand only for very simple cases, e.g. two-sample comparison.

You'll need some preliminary data, or some basic summary statistics, or at least some rough estimates, in order to perform a power analysis.
