NKX3.1短发卡RNA表达载体对NKX3.1基因表达的抑制作用

摘　要：目的研究同源框基因NKX3.1短发卡RNA(shRNA)表达载体在前列腺癌LNCaP细胞中对目的基因的沉默作用,为研究NKX3.1基因功能建立平台.方法构建针对NKX3.1的shRNA质粒载体(pU6/NKX3.1-1和pU6/NKX3.1-2),并观察其介导LNCaP细胞中NKX3.1表达沉默的效果以及NKX3.1表达沉默对LNCaP细胞生长的影响.结果构建的质粒pU6/NKX3.1-1和pU6/NKX3.1-2能够有效地抑制NKX3.1的表达,使NKX3.1mRNA和蛋白表达量显著下降,其中pU6/NKX3.1-2的效应高于pU6/NKX3.1-1,分别达到83.7%和72.1%.NKX3.1表达沉默能够使LNCaP细胞生长加快,细胞形态无明显改变.结论成功构建NKX3.1特异性shRNA真核表达载体,使LNCaP中的NKX3.1表达下调,初步证明NKX3.1在LNCaP细胞生长中起非常重要的作用,为进一步深入研究NKX3.1基因的功能提供实验基础.
