There is no conclusive answer to the enzyme responsible for the thiolester linkage of agrD although it was suggested by some authors that agrB protein might be. Currently, no experimental evidence is available for this claim.
Again, the requirement of thiolactone ring to the effective binding between agrD and agrC has not been tested directly. However, to change the downstream histidine protein kinase function of agrC,either activation or cross-inhibition, the thiolester ring of agrD is required.
